چکیده

Despite the widespread use of barbiturates and their clear potential for abuse, little is known of their neurochemical mechanisms of action. Our purpose is to review the neurochemical effects of acute and chronic barbiturate administration, with particular emphasis on the relationship between neurochemical changes and the development of tolerance and dependence. The electrophysiological effects of barbiturates have been reviewed recently (1-3), and Okamoto (4) has compared the neurophysiological and neurochemical effects of barbiturates to the effects of alcohol. The clinical charac.teristics of barbiturate abuse have been reported by Wesson & Smith (5), and the membrane actions of barbiturates and other anesthetics have been reviewed by Seeman (6) and Roth (7).

Barbiturate-induced alterations in the kinetic parameters of slow outward current in Aplysia giant neurons.

Regulation of spike frequency adaptation has been postulated to be a neuronal mechanism of action of the barbiturates (Zbicz and Wilson, 1981). A barbiturate sensitive slow outward current is present in Aplysia giant neurons, and this current is very effective at regulating adaptation. In the current study a quantitative method of determining the effect of barbiturates was developed using a sin...

SYNTHESIS OF NEW PHASE- 1 METABOLITES OF TWO BARBITURATES

The new 4'- hydroxy and 3'- aldehyde phase- 1 metabolites of butobarbitone and pentobarbitone are fully synthesized for the first time. These metabolites were used to idenfity and quantify the natural metabolites in human urine after single and multiple doses of these drugs. The synthesized metobolites have spectral characterizations (IR, NMR, U.V. and mass spectroscopy) consisted with the ...

Efficacy of different antiemetics with different mechanism of action on xylazine induced emesis in cats

Xylazine hydrochloride, α2 adrenoceptor agonist, is an analgesic, sedative, tranquilizer, and muscle relaxant agent in veterinary medicine which is mostly used as in pre-anesthesia in veterinary medicine. One of the main adverse effects of xylazine which limits application of this medication in small animal veterinary practice (mostly in cats) is nausea and vomiting which can end up with aspira...

How theories evolved concerning the mechanism of action of barbiturates.

The barbiturate phenobarbital has been in use in the treatment of epilepsy for 100 years. It has long been recognized that barbiturates act by prolonging and potentiating the action of γ-aminobutyric acid (GABA) on GABA(A) receptors and at higher concentrations directly activating the receptors. A large body of data supports the concept that GABA(A) receptors are the primary central nervous sys...

Mechanisms of action of currently prescribed and newly developed antiepileptic drugs.

Clinically available antiepileptic drugs (AEDs) decrease membrane excitability by interacting with neurotransmitter receptors or ion channels. AEDs developed before 1980 appear to act on sodium (Na) channels, gamma-aminobutyric acid A (GABAA) receptors, or calcium (Ca) channels. Benzodiazepines and barbiturates enhance GABAA-receptor-mediated inhibition. Phenytoin, carbamazepine and, possibly, ...
